生血胶囊对免疫介导的再生障碍性贫血小鼠骨髓Cyclin D_3表达的影响

摘　　要：目的　探讨生血胶囊对免疫介导的再生障碍性贫血 (再障 )小鼠骨髓增殖及细胞周期蛋白D3(CyclinD3)表达的影响。方法　建立免疫介导的再障模型。造模当日开始胃饲生血胶囊水提液 ,每次 0 .1mL/1 0 g体重 ,2次 /d ,连续 9d ,以生血丸为对照。第 1 0天处死动物 ,采用流式细胞仪检测小鼠骨髓有核细胞 (BMNC)G0 /G1 期细胞百分率 ,免疫组织化学超敏S AP法检测股骨中段骨髓CyclinD3表达水平。结果　生血胶囊组与再障模型组比较 ,G0 /G1 期细胞百分率显著降低 (P <0 .0 1 ) ,而CyclinD3表达水平明显升高 (P <0 .0 1 ) ,与生血丸组比较各项指标均无显著差异 (P >0 .0 5 )。结论　生血胶囊能提高免疫介导的再障小鼠骨髓CyclinD3的表达水平 ,促进骨髓造血细胞由G0 /G1 期进入增殖期 ,其作用强度与生血丸相当。Objective To study the influence of Blood Replenishing Capsule(BRC) on expression of Cyclin D 3 and the proliferation of hematopoietic cells in bone marrow of immune induced aplastic anemia (AA) mice. Methods After establishing 32 rats AA models,we fed the rats with BRC, 0.1?mL/10g(w), twice a day for 9?d.Blood Replenishing Pill 1(BRP 1) was selected as control.All rats were killed in 10?d after experiment.The percentage of cells in G 0/G 1 in bone marrow was analysed by flow cytometor analysis system,and the expression level of Cyclin D 3 was determined by immunohistochemical method.Results Percentage of cells in stage G 0/G 1 in bone marrow was lower than that in group AA( P <0.01), and the expression level of Cyclin D 3 in group BRC was significantly higher than that in group AA ( P <0.01). No obvious difference was found between group BRC and group BRP 1 ( P >0.05).Conclusion BRC could promote the proliferation of bone marrow of rats with AA by raising the expression level of Cyclin D 3. The effect of BRC was similar to that of BRP 1.
