摘  要:目的 探讨牵张刺激引起心肌细胞 β- MHC基因重新表达的机制。方法 在硅橡胶模上培养心肌细胞并施加牵张刺激 ,用免疫组织化学方法观测 NF- κB的激活情况 ,并用图像分析法检测 NF- κB在激活与未激活状态下 β- MHC蛋白的含量。结果  1心肌细胞受到应变为 2 0 %的牵张刺激 5小时后 ,NF- κB被显著激活 ,从胞浆移位入胞核。 2牵张组心肌细胞与对照组、NAC(N-乙酰半胱氨酸 )组和牵张 + NAC组心肌细胞相比 ,β- MHC含量明显增加 ,其它 3组 β- MHC含量无明显差异。结论  NF- κB信号传导系统参与了牵张诱导的 β- MHC基因表达的调控。Objective To determine the mechanism of reexpression of β MHC gene caused by the stimulation of stretch.Methods The specimens were taken from the 1 ,2 and 3 day old SD rats.Immunohistochemical method was applied to observe the activation of NF κB in stretched cardiac myocytes cultured on the silicon membrane, and then with the system of image analysis, the β MHC protein content was detected in four groups where NF κB was activated or not activated, respectively. Results NF κB was activated significantly by stretching cardiac myocytes for five hours. The β MHC protein content in stretched myocytes increased significantly, compared with that in the control myocytes, NAC treated myocytes, and stretch treated myocytes in the presence of NAC.Conclusion The above data demonstrate that NF κB signaling system is involved in regulating the stretch induced gene expression of β MHC.
