应用PCR/SSCP分析恙虫病东方体分离株基因型的研究

摘　　要：目的　对分离自辽宁、吉林地区的 6株恙虫病东方体目的基因进行分析研究 ,并与国际参考株进行比较。方法　经PCR/SSCP检测 ,呈现出 2种不同的单链构象图谱 :HCAa92 0 2株、HCAp92 0 3株及HCM95 0 1株图谱与Gilliam参考株相同 ,而KDCt940 2株、KDCp940 3株及KDCt940 4株的图谱则与Karp参考株相同。结果　辽宁、吉林地区恙虫病东方体分离株至少存在 2种型别。结论　应用PCR/SSCP方法分析恙虫病东方体基因型 ,简便、经济、省时、结果准确。Objective Using PCR/SSCP to analyse the gene type of the six Orientia stustugamushi(Ot) strains isolated from Liaoning and Jilin areas and making a comparision between the isolated strains of Ot and the international reference strains.Methods The six strains from Liaoning and Jilin appeared two different kinds spectrum of SSCP:The strains HCAa9202,HCAp9203 and HCM9501 had the same gene type with the reference strain Gilliam;strains KDCt9402,KDCp9403 and KDCt9404 had the same gene type with reference strain Karp.Result Ot strains isolated from Liaoning and Jilin areas had at least two different types identified by PCR/SSCP analysis.Conclusion The method of PCR/SSCP was proved to be simple,economic,time-saving and accurate.
