摘 要:目的 :观察去卵巢 ( OVX)成年雌鼠的认知功能、脑内神经生化学改变及补充雌激素的影响。方法 :通过跳台法测试动物被动及主动回避反应 ,神经生化学手段测定相关酶及受体活性。结果 :OVX组大鼠的学习记忆能力明显下降 ;大脑皮层和海马部位的胆碱乙酰化转移酶 ( Ch AT)活性显著低下 ;大脑皮层胆碱能 M受体和 N-甲基 -D-天门冬氨酸 ( NMDA)受体结合活性明显降低而单胺氧化酶 B( MAO-B)活性增加。经补充苯甲酸雌二醇 ( EB)或植物雌激素 NH1 9966周后 ,其学习记忆明显改善 ,上述生化指标均接近正常。结论 :推测雌激素和 NH1 996可通过影响中枢胆碱能 。To observe the effects of estrogen and NH1996 on cognition and neurochemical changes including central cholinergic, glutamatergic and monoaminergic systems in brain of adult ovariectomized (OVX) rats. Methods: Behavioral study and neurochemical study were used. Results: In OVX group, the behavioral study examing passive and active avoidance tasks showed that learning and memory capability was weakened. Neurochemical study showed that choline acetyltransferase (ChAT) activity in the cerebral cortex and hippocampus decreased significantly; the specific binding of 3H QNB for m ACh receptor(m AChR) activity and 3H MK801 for N methyl D aspartate receptor (NMDAR) in the cerebral cotrex decreased significantly; while the activity of monoamine oxidase B(MAO B) increased. After treating with EB or NH1996 at a dosage of 200 μg per rat, im, once every three days, for six weeks, all changes mentioned above were reversed. Conclusions: EB and NH1996 take part in the regulation of learning and memory through cholinergic, glutamatergic and monoaminergic mechamisms.
