摘 要:目的 :寻找小肠移植排斥反应的标志物 ,研究排斥反应的分子生物学机制。 方法 :利用显微外科技术建立小鼠节段性小肠移植模型 ,采用逆转录定量聚合酶链式反应技术检测受体淋巴细胞中白细胞介素 2受体 (IL - 2R)α链编码基因功能区的表达水平 ,并对反应产物进行DNA序列测定。 结果 :小肠移植术后 72h ,异系移植组 (津白Ⅱ号→BALB/c)受体鼠淋巴细胞IL - 2Rα链功能区的转录水平 (0 85± 0 17)显著高于同系移植组 (0 6 0± 0 0 6 )和假手术组 (0 5 7± 0 2 1)。DNA序列测定证实PCR反应产物与IL - 2Rα链功能区编码基因 5’端序列相同。 结论 :小肠移植早期IL - 2Rα链编码基因mRNA的高表达是小肠移植排斥反应的主要启动因素和早期标志物 。To search for the rejection marker of SBT and to study the biomolecular mechanism of rejection Methods:The segmental SBT model in mice was established with microsurgical procedure.The reverse transcription level of the gene coding Interleukin 2 receptor(IL-2R)α chain in recipients' lymphocyte was quantitatively assayed with Quantitative Reverse Transcription Polymerase Chain Reaction(QRT-PCR)and the sequence of the target gene fragment was determined.Results:At postoperative 72 hours,the mRNA reverse transcrption levels of functional domain of IL-2Rα chain in allograft group(Jinbai No.II→BALC/c)(0.85±0.17)was higher than those in isograft group(0.60±0.06)and control group(0.57±0.17).The 5' sequence of target gene fragment was proved to be the same as the results from the reported.Conclusion:In recipients,the higher significantly transcription level or IL-2Rα chain after SDT in rejective group indicated that the express of gene coding IL-2Rα chain was an important promoter and early gene marker of rejection of STB.The sequence of target gene fragment demonstrated its specialty.
