摘 要:目的 探讨血脂康的非调脂功能。方法 体外培养的巨噬细胞与血脂康或血脂康加甲羟戊酸 (羟甲基戊二酸单酰辅酶A还原酶产物 )孵育 ,凝胶酶谱法检测上述细胞培养基中基质金属蛋白酶 (matrixmetallopro teinases,MMPs)活性 ,WesternBlot方法鉴定MMPs类型。结果 血脂康在体外抑制巨噬细胞分泌MMP 2的活性 ,这种抑制作用呈药物浓度依赖。结论 血脂康在体外抑制巨噬细胞分泌的基质金属蛋白酶Objective To study the non lipid regulation function of Xuezhikang, a kind of Statins lipid regulation drug(β hydroxy β methylglutary1 CoA reductase). Methods The conditioned media was generated by incubating the macrophage for 24 hours at 37℃ with MEM supplemented with 10% FBS and different concentrations of Xuezhikang in the absence or presence of mevalonate. The activity of matrix metalloproteinases (MMPs) secreted by cultured Syrian Golden Hamster macrophage was measured using enzymography and characterized by Western Blot. Results MMP 2 was found to be inhibited by Xuezhikang in a dose dependent manner and the effect of inhibition could be reversed by adding exogenous mevalonate. Conclusion Xuezhikang has the inhibitory effect on MMP 2 secretion by macrophage.
