摘  要:目的 探讨冻伤后组织超微结构的变化 ,为冻伤诊断和治疗提供参考依据。方法 用大耳白家兔进行复制Ⅰ~Ⅳ度冻伤模型 ,在冻伤后 12 ,2 4,48,72h时分别采取冻伤组织标本 ,常规电镜制样 ,CH10型透射电镜观察其冻伤后组织超微结构的变化。结果 ①轻度冻伤 (Ⅰ~Ⅱ度 ) 12~ 2 4h时上皮间质水肿 ,肌肉亦有水肿 ,48~ 72h时上皮细胞有退行性变 ,肌肉也有退行性变 ,个别肌束的Z线、H带不清偶见肌丝紊乱。②重度冻伤 (Ⅲ~Ⅳ度 ) 12~ 2 4h时上皮细胞有不规则的排列 ,线粒体分布不一 ,Z线、H带部分区域结构不清 ,线粒体偶见空化 ,48~ 72h时上皮细胞排列紊乱 ,仅见细胞轮廓 ,细胞器极少 ,线粒体大片空化 ,肌束紊乱、断裂 ,Z线、H带消失。③重度冻伤比轻度冻伤其超微结构改变明显。结论 随着冻伤程度的加重 ,其组织超微结构破坏越严重。Objective The aim of this study was to investigate the ultrastructure changes after frostbite,and to provide the basis for diagnosis and treatment of the frostbite.Methods Frostbite model on rabbit foot was duplicated.Specimens at 12,24,48,and 72 hours after frostbite were collected,and the ultrastructure changes were observed with a CH10 model electron\|transmission microscope.Results (1)Oedema occurred in dermal intestine and muscle at 12~ 24 h after light frostbite(Ⅰ~Ⅱ degree).Degenerative changes in dermal cells and muscle occurred .Z line and H tape in individual muscle bundle are obscured,occasionally muscle bundle arranged irregularly at 48~ 72 h after frostbite.(2)In case of severe frostbite(Ⅲ~Ⅳdegree),the arrangement of dermal cells was irregular,and mitochondria was unevenly distributed.The structure of Z line and H tape was obscured.Vacuoles in mitochondria occurred at 12~ 24 h after frostbite.Dermal cells arranged disorderly,and mitochondria vacuolated extensively.Cell organ was very rare,only cell outline could be seen.Muscle bundles were disordered and broken.Z line and H tapes disappeared at 48~ 72 h after frostbite.(3)Ultrastructure changes in severe frostbite showed much obvious than those shown in light frostbite.Conclusions As frostbite aggravated,the damages in ultrastructure increased.
