摘  要:目的 探讨中国维吾尔族、彝族人群δ 氨基 γ 酮戊酸脱水酶 (ALAD)基因多态性和等位基因的分布规律。方法 采用聚合酶链式反应 (PCR)和MspI限制性片断长度多态性 (RFPL)对 2 14名新疆维吾尔族人和 144名云南彝族人的ALAD基因多态性进行分析。结果 在维吾尔族人群中ALAD1 1纯合子基因型占 82 .71% ,ALAD1 2杂合子占 15 .89% ,ALAD2 2纯合子占 1.40 % ;ALAD1和ALAD2 等位基因频率分别为 90 .6 5 %和 9.35 %。在彝族人群中ALAD1 1纯合子基因型占 96 .5 3% ,ALAD1 2杂合子占 2 .87% ,而ALAD2 2纯合子仅占 0 .6 9% ;ALAD1和ALAD2 等位基因频率分别为97.92 %和 2 .0 8%。结论 中国维吾尔族人群的ALAD2 等位基因频率分布与文献报道的欧洲白人ALAD2 等位基因频率分布相近 ,而彝族人群的ALAD2 等位基因频率分布与文献报道的亚洲人群相近。Objective To explore the genetic polymorphism and the distribution of allelic frequencies of δ aminolevulinic acid dehydratase(ALAD)(E.C.4.2.1.24) in Uighur and Yi population in China. Methods The ALAD genotyping was determined by polymerase chain reaction(PCR) and MspI restriction fragment length polymorphic techniques in 214 unrelated Uighur individuals from Xinjiang province and in 144 unrelated Yi individuals from Yunnan province. Results The percentage of ALAD1 1 homozygote was 82.71%,ALAD1 2 heterozygotes was 15.89% and ALAD2 2 homozygote was 1.40% in Uighur subjects.The frequencies of the allele ALAD 1 and ALAD 2 were 90.65% and 9.35%,respectively.The percentage of ALAD1 1 homozygote was 96.53%, ALAD1 2 heterozygotes was 2.78% and ALAD2 2 homozygote was 0.69% in Yi subjects.The frequencies of the allele ALAD 1 and ALAD 2 were 97.92% and 2.08%,respectively. Conclusion The frequency of the ALAD 2 allele in Uighur was similar to that in Caucasian population.The frequency of the ALAD 2 allele in Yi was similar to that in Asian population.
