摘 要:目的 通过观察脊髓损伤后中枢下丘脑室旁核、视上核c -fos样蛋白免疫反应了解中枢与脊髓损伤之间的关系。 方法 成年大鼠 5 6只 ,体重 (2 70± 2 0 )g,分为脊髓损伤组 (2 0只 )、假手术组 (2 0只 )、正常对照组 (16只 )。损伤模型为双后肢不全瘫。伤后 1,6 ,12 ,2 4h对下丘脑室旁核、视上核组织进行冠状面切片 ,运用c -fos漂片免疫组化方式对阳性细胞计数。 结果 脊髓损伤后不同时间组的下丘脑室旁核、视上核均有明显的c -fos阳性表达。伤后 1,6 ,12 ,2 4h室旁核c-fos阳性表达细胞数与假手术组、正常对照组比较 ,差异有非常显著性意义 (P <0 .0 1) ;伤后 12 ,2 4h视上核c -fos阳性表达细胞与假手术组、正常对照组比较 ,差异有非常显著性意义 (P <0 .0 1)。 结论 脊髓损伤影响正常神经信号在大脑中枢传导 ,下丘脑室旁核、视上核可产生明显反应。Objective To study c-fos expression in hypothalamic pa raventricular nucleus (PVN) and supraoptic nucleus (SON) after thoracic spinal cord injury. Methods A total of 56 adult male rats weighing (2 70±20) g were divided into injury group (Group A), false operation group (Group B) and control group. Rat spinal cord was injured by Allen's method (80·cm). A ll animals were sacrificed 1, 6, 12 and 24 hours respectively after surgery. The sections of the rat brain were subjected to c-fos immunohistochemical reaction with ABC method. Results One hour and 6, 12, 24 hours after injury, c-fos positiv e cell e xpression of PVN was significant compared with that in Group B and control grou p ( P <0.01); 12, 24 hours after injury, c-fos positive cell expression of SO N was significant compared with Group B and control group ( P <0.01). Conclusions The present findings suggest that spinal cord injury affects nervous signal conduction and induces significant reaction in ret ated nuclei in the brain.
