摘 要:本文以20期鸡胚水晶体为实验模型,用原位注射的方法,研究了乌本苷干扰生长控制的可能性。结果发现,所测试的3种浓度(0.1,0.5和2.0μmol/L)的乌本苷双蒸水溶液,均可导致水晶体明显增大。同时,乌本苷处理后,水晶体纤维细胞数也明显增多。但单纯双蒸水的注射,并不引起水晶体的明显增大。正常鸡胚左右侧水晶体之间,亦无明显的大小差异。以上结果证明,乌本苷确可干扰生长控制,其机制可能是乌本苷干扰了连接通讯,从而造成生长失控。本文还报道,如果溶剂中含有蛋白质等成分,可降低乌本苷的促增生效果。It has been reported that ouabain interferes with junctional communication. We have known that junctional communication influences growth control. Therefore, ouabain would be expected to interfere with growth control. The study reported here attempts to demonstrate this possibility.Ouabain at a concentration of 0.1 μM, 0.5 μM or 2.0 μM in redistilled water was injected into the right eye of chicken embryos at the 20th stage; the opposite uninjected left eye was used for the control. The size of lenses was measured 24 hours after injection, and the difference in size between experimental and control groups of lenses was compared statistically. The data show that the lenses from injected eyes are significantly larger than control (table 1, P<0.05 or P<0.001). When the ouabain was dissolved in DMEM + 10% FCS, lenses injected with 1.0 μM ouabain were significantly larger than uninjected control, but lenses injected with 0.1 μM ouabain were not. Although low concentrations administered in DMEM +10% FCS are less effective than in water, excess growth is apparent at higher concentrations.To rule out effects of redistilled water and of DMEM + 10% FCS, we carried out the injection with redistilled water or DMEM +10% ECS. No significant difference could be detected in any of these cases (table 2, P>0.05). The two lenses of an untreated chicken embryo are also not significantly different in size (table 3, P>0.05).Microscopic morphology has also been observed, no visible damage could be found in the structure of lenses from eyes injected with ouabain (figs 3 and 4). Serial sections of paraffin-embedded lenses show that the number of fiber cells increased significantly in experimental samples treated with ouabain at a concentration of 0.1 μM(table 4, P<0.05).These data demonstrate that ouabain may indeed interfere with growth control, resultin gin overgrowth of lenses, The mechanism of lens overgrowth caused by ouabain has been considered.
