摘  要:参照Mroz的方法制备P——物质抗原,用六只家兔免疫制备P物质抗体。经RIA测定N.10SP抗体漓度为1:150,000——180,000.用Scatchard作图求得抗体亲和常数Kd=3.5×10^(10)L/M。以免疫细胞化学染色检验抗体之特异性灵敏性。其工作稀释浓度IF法为1:4000~8000,PAP及ABC法为1:10,000~20,000,染色背景低。P物质免疫反应神经纤维在大鼠及豚鼠的脊髓、胃肠道、胰、肾上腺等部位之形态与分布和文献报导相似。实验表明,自制N.10SP抗体可适用于放射免疫测定和免疫细胞化学染色。Refer to the Morz's method to preparate SP antigen, The antibody was raised in six rabbits against synthetic SP, The RIA titre of N.Io SP antibody was 1: 150, 000-180, 000, The average affinity constant of the antibody was kd=3.5×10^(10) L/M.as determined by Scatchard plot analysis. Immunocytochemical techniques were used to determine the sensitivity and specificely of N. IO SP antibody, The working dilution was Ⅰ: 4000-8000 in IF method ; 1: 10, 000-20000 in PAP and ABC methods,The stainings were with a very low background.The distribution and morphology of SP-immunoreactive nerve fibers in the spinal cord, gastrointestinal tract, Pancrease and adrenal gland etc.of rat and guinea-pig stained with N.IO SP antibody were similar to other laboratory's reports. It is showed that N.IO SP antibody was suitable for immunocytochemistry and radioimmunoassay studies.
