用免疫组织化学法检测自制NPY抗体的特异性和敏感性

摘　　要：本实验用戊二醛作偶联剂,将神经肽Y(NPY)偶联到牛血清白蛋白(BSA),制成NPY-BSA免疫原复合物。用它免疫新西兰大白兔,成功地制备了兔抗NPY抗血清。经免疫组織化学检测,其最高使用效价为1:1000～1:2000。替代试验和吸收试验均呈阴性反应。该抗血清能显示胃肠血管壁和脑底动脉管壁的神经纤维以及大鼠脑内的神经纤维及神经元胞体。它们的分布特点与进口NPY所显示的相一致。以上结果表明,该抗血清是对NPY特异性的且有较高使用效价。Synthetic Neuropeptide Y(NPY) was coupled to bovine serum albumin (BSA) by glu(?)araldehyde. Antiserum was raised by immunising New Zealand rabbits with the NPY-BSA complex. The antiserum was tested with the immunohystochemistry ABC technique. It showed positive nerve fibres around the blood vessels in the rat gastrointestinal tract and the cerebral arteries. Positive nerve fibres and neuronal cell bodies were also found in the rat brain.The working dilution was 1:1, 000-1:2, 000.The pattern of the positive staining obtained with the antiserum was the same as that with a specific antiserum to NPY.The substitution and absorption tests were negative. The results suggeste that the raised antiserum is a specific antiserum to NPY with a high titre.
