摘  要:目的 建立准确检测载脂蛋白CⅡ (apoCⅡ )微卫星DNA(TG)n(AG)m的方法。方法 采用套式聚合酶链反应扩增样品DNA ,将扩增产物上样于变性聚丙烯酰胺凝胶 ,高压电泳 ,凝胶银染后按等位基因梯阶标准判定样品基因型 ;对等位基因(TG) 2 0 (AG) 8进行测序。结果  (TG)n(AG)m位点 ,共检出 12个等位基因 ( 17、18、2 6~ 3 5 ) ,3 6个基因型。 (AG)m位点 ,共检出4个等位基因 ( 6~ 9) ,7个基因型。DNA测序确证该等位基因为 (TG) 2 0 (AG) 8。结论 该法准确、快速、简便 ,适用于科研及大规模人群检查。Objective\ To establish an accurate method for detecting apolipoprotein CⅡ microsatellite DNA(TG)n(AG)m Methods \ The DNA samples were detected by nested polymerase chain reaction combing high voltage denaturing polyacrylamide gel electrophoresis and sliver stain The lengths of PCR products were determined by the allelic ladders of the two loci On the other hand,the allele of (TG) 20 (AG) 8 was sequenced Results\ 36 different genotypes and 12 different alleles(17?18?26~35)of apolipoprotein CⅡmicrosatellite DNA(TG)n(AG)m locus were detected in 231 healthy individuals,while 7 different genotypes and 4 different alleles(6~9)of (AG)m locus were detected DNA sequence confirmed that the allele was(TG) 20 (AG) 8 Conclusion\ The method is accurate,rapid,and simple It can be applied to science research and large scale population investigation
