青岛地区TTV抗体阳性人群的病毒基因检测及序列分析

摘　　要：①目的　了解经血传播病毒 (TTV)在青岛地区的基因变异情况。②方法　选择TTV抗体阳性的标本 ,用巢式PCR技术扩增血清中的DNA特定片段 ,纯化后与载体连接并转入大肠杆菌JM10 9,提取阳性克隆的重组质粒测序并与TTV的已知序列相比较。③结果　在选择的 2 0例标本中 ,15例扩增出TTVDNA ,对其中 1株测序后与已知序列AB0 0 8394比较 ,发现碱基变异率为 13.3 % .Objective\ To investigate the variation of TTV gene in Qingdao region. \ Methods\ Specimens of positive serum of TTV antibody were collected and the specific fragments of TTV gene were amplified using nested PCR, the products were purified and connected with T vectors, and then cloned into E.coli JM109, the recombinant plasmid DNA sequence was extracted and compared with AB008394.\ Results\ Fifteen fragments were amplified among 20 samples, it was found that the variation rate of base was 13.3% as compared with that of Ab008394.\ Conclusion\ There are variant TTV in Qingdao area.
