摘 要:为了探讨骨必康对绝经后骨质疏松症的防治效果及其作用机理 ,选用 60只 8月龄 SD雌性大鼠 ,随机分为假手术组(SHAM)、模型组 (OVX)、尼尔雌醇组 (CEE3)、骨必康低剂量组 (GBK- L )和骨必康高剂量组 (GBK- H) ,每组 12只。假手术组不切除卵巢 ,其余各组均切除双侧卵巢 ,术后第 4天开始灌胃给药 ,12周后肝静脉取血 ,处死动物。分离血清和血浆 ,测定 S- AKP、S- BGP、S- Ca、S- P及血浆 ACP,摘取 L3 椎体及左侧股骨测定 BMC、BMD。结果显示 ,与模型组比较 ,骨必康低、高剂量组均能明显提高 L3 椎体及左侧股骨 BMD(P<0 .0 5~ 0 .0 1) ,且高剂量组能明显降低血浆 ACP(P<0 .0 0 1) ;与假手术组比较 ,中药组 S-BGP、S- AKP含量明显升高 (P<0 .0 5~ 0 .0 0 1)。提示骨必康对绝经后骨质疏松症具有肯定的防治作用 ,其作用机制在于抑制骨吸收 ,促进骨形成 。To explore the preventive effects and the mechanisms of Gubikang on the treatment of postmenopausal osteoporosis, sixty 8-month SD female rats were divided into 5 equal groups: pseudo operation group, model group, nylestriol group, Gubikang in low dosage group, and Gubikang in high dosage group. The abdomen of the first group was only made an incision, the other 4 groups were also made an abdominal incision and ovariectomized bilaterally, then they were treated by water, water, nylestriol, Gubikang in low dosage, Gubikang in high dosage since the 4th day after operation. Twelve weeks later each rat was drawn blood from the liver vessel, then was killed. The serum and plasma were isolated to measure serum alkaline phosphatase (S-AKP), serum osteocalcin (S-BGP), serum-calcium (S-Ca), serum phosphorus (S-P) and plasma acid phosphatase (ACP), the BMC and BMD of the 3 rd lumbar vertebra and of the left femur were also measured. The results showed that compared with the model group, the BMD of L 3 and the left femur in both Gubikang groups could significantly be increased (P<0.05~0.01), and the plasma ACP in the high dosage group was decreased significantly (P<0.001); compared with the pseudo operation group, the level of S-BGP, S-AKP in both Gubikang groups could be enhanced evidently, suggesting that Gubikang has definite preventive effects on postmenopausal osteoporosis, the mechanisms of Gubikang on the treatment of postmenopausal osteoporosis are the inhibition of the bone resorption, the promotion of the osteogenesis, and the prevention of the bone mass loss.
