新伤续断汤对骨折愈合中胶原影响的实验研究

摘　　要：目的 :探讨中药新伤续断汤对骨折愈合中胶原代谢的影响。方法 :成年 SD雄性大鼠 2 4只 ,于胫骨上段造成 2 mm的骨缺损。术后随机分为治疗组和对照组 ,分别给予新伤续断汤和生理盐水 ,并于 1 0、2 0、3 0天后处死。标本通过组织学及免疫组化染色进行观察。结果 :组织形态学显示治疗组血肿吸收、机化较快 ,软骨性骨痂出现早 ,骨化提前。免疫组化染色显示 :术后 1 0天治疗组 、 型胶原染色深于对照组 ,2 0天治疗组 型胶原染色明显强于对照组 ,3 0天治疗组 型胶原染色仍较深 ,而且排列规则。结论 :新伤续断汤能加速骨折愈合。其机制在于其能够改善局部血液循环 ,促进血肿的吸收、机化、加速各型胶原的合成 ,改善胶原的结构排列。Objective:To investigate the effects of Xin Shang Xu Duan Tang on collagenⅠ?Ⅱ?Ⅲ metabolism during fracture healing.Methods:The bone defects of rats proximal tibial in both limb were performed, and they were divided into 2 groups randomly.The experimental group was killed at 10,20,30 days respectively after operation.The specimens(callus) were observed with histological and immuno-histochemical examination.Results:Histological examination showed that the absorption and organization of hematoma,the appearance of cartilage callus and endochondral ossification in experimental group were earlier than those in control group.Immunohistochemical study showed that the intensity of collagen Ⅱ,Ⅲ stain in experimental group was darker than the control group at 10 days, and the intensity of collagen Ⅰstain in experimental group was significantly darker than the control group at 20 days, and the intensity of collagen Ⅰ stain in experimental group at 30 days was still darker than that at 20 days, furthermore the arrangement of collagen was regular.Conclusion:Xin Shang Xu Duan Tang can increase the collagen synthesis and improve the arrangement of collagen structure.
