摘  要:目的 :探讨耐热乳糖酶产生菌的筛选方法及其产酶条件。方法 :利用含有 X-gal平板培养基 ,于 6 0℃划线培养 ,筛选出耐热乳糖酶产生菌 ,并观测了培养温度、培养时间、培养基初始 p H及乳糖浓度对菌株生长及产酶条件的影响。结果 :筛选出含耐热乳糖酶菌株 ,该菌株的最适产酶条件分别为培养温度 (6 0± 1 )℃ ,培养时间 1 8~ 2 1 h,培养基起始 p H7.0~ 8.0 ,培养基乳糖浓度 41 .7mmol/ L。结论 :在含有 X-gal培养基进行划线培养是筛选产乳糖酶菌株的有效方法 ,同时该菌株具有嗜热性。Objective:To screen thermostable lactase producing strain of Thermus sp and optimize its culture condition.Methods:Thermus strains producing thermostable lactase were screend in culture medium containing X gal.The conditions for lactase production were studied,such as incubation temperature,culture time,initial pH and lactose concentration in culture medium. Results: A strain of thermus for producing thermostable lactase was selected.The optimal conditions for lactase production were obtained:initial pH was 7.0~8.0; incubation temperature was (60±1)℃;culture time was 18~21 h and concentration of the lactose was 41.7 mmol/L.Conclusion:Thermus strains producing thermostable lactase can be effectively obtained by culture with medium containing X gal.
