摘  要:采用大鼠局灶性脑缺血再灌流实验动物模型 ,观察当归对于脑缺血损伤的保护作用。采用线栓法制作单侧大鼠大脑中动脉栓塞模型 ,缺血 2h ,再灌 4 8h。当归治疗组缺血再灌后静脉滴注 5 0 %当归注射液 3 5~ 4ml。原位末端标记法观察细胞凋亡并计算凋亡指数 ,免疫组化观察微管相关蛋白 (MAP 2 )的表达。结果显示 ,当归治疗组较缺血再灌组脑缺血组织细胞凋亡明显减少 ,凋亡率下降 ,当归治疗组MAP 2的表达明显恢复 ,接近对照组。结论 ,当归能减少脑缺血损伤后缺血区细胞凋亡的发生 ,促进微管相关蛋白 (MAP 2 )表达 ,这是当归对大鼠局灶性脑缺血损伤保护作用的可能机制之一。Objective:This expriment observed the protection of angelica on focal cerebral ischemia injury of rat brain.Methods:The embolism of Nylon thread produced animal models of the middle cerebral artery occlusion.After 2 hours of ischemia,the time of reperfusion was 48 hours.The group treated with angelica was injected angelica(iv,3.5-4ml) slowly after ischemia-reperfusion.Using immunohistochemical staining methods,the expression of microtubule-associated protein 2(MAP-2) was observed.And terminal deoxynucleotidyl-trallsferase(TDT)mediated DUTP-biotin nick end labeling(TUNEL) method was applied to detect apoptosis.Results:Apoptosis reduced evidently in the group treated with angelica.The expression of MAP-2 in the ischemia region of group treated with angelica enhanced,close to the control group.Conclusions:Angelica can reduce apoptosis and promote the expression of MAP-2.This may be one of the mechanism of the protection of angelica on focal cerebral ischemia injury of rat brain.
