摘 要:目的 研究全反式维甲酸对体外培养大鼠系膜细胞uPA和MMP 2表达的影响 ,探讨全反式维甲酸抗肾小球硬化的机制。方法 分别应用Northernblot、Westernblot和酶谱法检测全反式维甲酸对培养大鼠系膜细胞uPA和MMP 2表达的影响。结果 ATRA可促进系膜细胞uPA和MMP 2蛋白表达 ,提高其酶活性 ,并增强MMP 2mRNA表达。结论 ATRA可增强大鼠系膜细胞的uPA和MMP - 2表达 。Purpose: To study the effects of all-trans retinoic acid on the expression of uPA/MMP-2 in cultured rat mesangial cells, and to elucidate the mechanism by which all-trans retinoic acid prevents age-related progressive glomemular sclerosis. Methods: The mRNA, protein and enzymatic activity of uPA/MMP-2 were detected by Northern blot analysis, Western blot analysis and Zymography assay, respectively. Results: All-trans retinoic acid upregulated the expression of uPA/MMP-2 protein, enhanced the enzymatic activity of uPA/MMP-2. Thus all-trans retinoic acid elevated the level of MMP-2 mRNA in cultured mesangial cells. Conclusions: All-trans retinoic acid upregulated the expression of uPA/MMP-2 in cultured rat mesangial cells. It is valuable for explaining the role of all-trans retinoic acid in preventing age-related glomerulsclerosis.
