摘 要:目的 :观察造血干细胞表面粘附分子的变化在外周血干细胞动员中的作用。方法 :用双色免疫荧光法研究 15例恶性血液病患者经化疗 +G CSF动员外周血干细胞前后、骨髓和外周血CD34 + 细胞表面 β1整合素 (CD49d)、β2 整合素 (CD11a、CD11b)及L 选择素 (CD6 2L)的表达。结果 :①动员后第 7天CD34 + 细胞表面较动员前CD49d、CD11a、CD6 2L表达下降 (P <0 0 1) ,而CD11b无变化 (P >0 0 5 )。②外周血CD34 + 细胞表达CD49d、CD11a、CD11b、CD6 2L较骨髓低 (P <0 0 5 )。③外周血CD34 + 细胞绝大多数处于G0 /G1期 ,该期的干细胞CD49d的密度低于S +G2 /M期。结论 :化疗 +G CSF通过粘附分子的变化而使造血干细胞从骨髓进入外周血中。Objective:To study whether the mobilization process is associated with change in expression of adhesion molecules on CD34 +cells Methods:Two colour fluorescence analysis was used to study the expression of adhesion molecule CD62L,CD49d,CD11a,CD11b on CD34 + cells of peripheral blood stem cells(PBSC) before and after mobilizing with G CSF and cytotoxic chemotherapy in 15 cancer patients undergoing PBCST Results:① The expression of adhesion molecules on CD34 +cells revealed a significant reduction of CD11a, CD49d and CD62L at days 7 compared to the baseline level (P<0 01) ②The proportion of CD34 + cells expressing CD62L,CD49d tended to be reduced in leukocytapheresis products compared with bone marrow ③ Most circulating mobilized CD34 + cells are in the G1 phase ,CD34 + cells in the G0/G1 phase expressed lower levels of CD49d than those in the S+G2/M phase Conclusion:These changes appeared to be involved in the mobilization mechanism of hematopoietic progenitor cell from BM into PB after cytotoxic chemotherapy and G CSF
