摘 要:选取健康及患丛枝病泡桐 (Paulownia spp.)为材料 ,建立组织培养和植株再生系统 ,以茎段作为转化受体 ,诱导分化和生根的最佳激素组合分别是 MS+BA 4mg/L +NAA 0 .2mg/L和 1 /2 MS+KT0 .5mg/L+IBA0 .2 5mg/L,芽分化频率可达 2 2 .8%。健康和患病泡桐的茎段经农杆菌共培养 3d后 ,在附加 50 mg/L Km的选择分化培养基上培养 2 0 d左右再生出抗性芽 ,经培养、诱导生根 ,获得了转基因再生植株 ,建立了泡桐的遗传转化体系。PCR和 Southern杂交检测证明外源基因已整合到泡桐的基因组 ,标记基因 ITP 在再生植株中也得到表达。Healthy and infectious (infected by Paulownia Witches Broom) Paulownia was selected as the investigated material.It was appropriate for two kinds of Paulownia to select stem segments as transplant,differentiate on MS+BA 4 mg/L+NAA 0.2 mg/L and induce root on 1/2 MS+KT 0.5 mg/L+IBA 0.25 mg/L,the differentiation efficiency could up to 22.8%.Stem segments were infected with Agrobacteria tumefaciens LBA4404 containing plant expression vector p438PRSI,and co cultured for three days.Resistant shoots were obtained about 20 days on MS media supplemented with kanamycin 50 mg/L.Regenerated plants were grown up after shoot culture and root inducing.PCR and Southern analysis of the transformants revealed that foreign gene has been integrated into plant genomes.Expression of report gene NPTⅡ was confirmed in regenerated plants.The factors influencing transfor mation of Paulownia were also discussed.
