作为抗肿瘤药物靶向载体LDE的可行性研究

摘　　要：目的 :观察LDE ACM复合物对裸鼠胃癌SGC 790 1、NKM 45细胞株皮下移植肿瘤的抑制作用 ,了解LDE作为抗肿瘤药物靶向载体的可行性。方法 :采用密度梯度离心和SephadexG 5 0层析以及超声法制备LDE和LDE ACM复合物 ,建立裸鼠胃癌皮下移植肿瘤模型 ,观察LDE ACM对移植肿瘤的抑制作用。结果 :LDE ACM复合物可明显抑制裸鼠皮下移植瘤的生长 ,与其他实验对照组比较 ,LDE ACM组的瘤重明显减轻 ,抑瘤率明显增高 ,而外周白细胞计数无明显降低。结论 :LDE ACM复合物有较强的抑制肿瘤效应且副作用明显减轻 ;LDE作为抗肿瘤药物靶向载体是可行的。Objective:To study on the potential uses of LDE as a targeted delivery carrier of anti tumor drugs,and the inhibitive effets of LDE ACM complex on nude mice′s subcutaneous implanted tumor.Methods:LDE ACM complex was prepared by the methods of ultrasound,density gradient centrifugation and SephadexG 50 chromatography.The model of nude mice's implanted tumors was set up by subcutaneous injection of gastric cancer cell lines,SGC 7901 and NKM 45.The inhibitory effect of LDE ACM complex on the implanted tumors was observed.Results:There were less tumor weight and higher rate of tumor repression as well as less decrease of leukocyte in the group of given LDE ACM complex than that in other groups.Conclusion:LDE ACM complex has significant effect on tumor repression and fewer side effects.It can be used as targeted delivery carrier of anti tumor drugs.
