摘 要:目的比较瘢痕疙瘩不同部位成纤维细胞生物学活性,以进一步探讨瘢痕疙瘩形成发展机制.方法采用体外培养成纤维细胞技术对所有标本进行细胞培养,取6~8代细胞作为实验对象.噻唑蓝(MTT)比色法比较细胞接种后不同时间瘢痕疙瘩不同部位成纤维细胞的增殖活性和碘化丙啶(P1)染色、流式细胞仪比较它们在无血清培养条件下和在不同浓度Fas单克隆抗体(Fas mAb)作用下的细胞凋亡率.结果瘢痕疙瘩边缘成纤维细胞增殖活性高于中央,两者均高于正常皮肤(P<0.01);无血清培养24 h后,瘢痕疙瘩边缘及中央细胞凋亡率均有所增加,但两者之间无显著性差异(P>0.05);FasmAb作用下,瘢痕疙瘩边缘及中央细胞均不能正常凋亡,两者之间无显著性差异(p>0.05).结论瘢痕疙瘩不同部位成纤维细胞生物学活性不尽相同,瘢痕疙瘩边缘成纤维细胞增殖活性过高可导致其向周围不断增生并浸润生长.Objective To observe the proliferating and apoptotic behavior of the fibroblasts derived from different parts of keloid and explore the mechanism of keloid formation and progression. Method Keloid samples were obtained surgically from patients with pathologically confirmed keloids and cell culture was conducted to select the fibroblasts of 6 to 8 passages for subsequent experiments. The proliferating activity of the fibroblasts derived from different parts of the keloids was assessed by MTT color: matric method and the apoptotic rate determined by flow cytometry when they were exposed to serum- free medium and Fas mAb of different concentrations. Results The proliferating activity of fibroblasts derived from the edges of the keloids was stronger than that of the fibroblasts from the central part of the keloids which in turn was stronger than that of normal skin (P<0.01). However, when exposed to serum-free medium and Fas mAb, the fibroblasts responded similarly in terms of apoptosis regardless of the region where they were taken (P>0.05). Conclusion The biological behavior of the fibroblasts derived from different parts of keloid may differ, and the fibroblasts on the edge of the keloids possess strong proliferating activity that may induce the overgrowth and infiltration of keloids.
