9V型肺炎球菌免疫血清的制备及应用

摘　　要：目的　制备 9V型肺炎球菌免疫血清 ,建立一套制备工艺 ,继续研制其他型的血清 ,为实验室鉴定肺炎球菌多糖疫苗的特异性所使用。方法　将 9V型菌株培养后制成死菌抗原 ,免疫家兔所得血清 ,加 0 0 1%硫柳汞 ,除菌过滤 ,无菌分装。结果　血清免疫抗原的试管凝集效价为 32 0X ,血清多糖抗原的效价为 16X ,原倍血清和不同浓度的多糖抗原免疫双扩试验检测多糖的灵敏≥ 2 5 μg/ml ,不同浓度的血清和 6 0 0 0 μg/ml多糖抗原的免疫双扩试验的效价为 2X ,免疫双扩试验证实本实验制备的 9V型肺炎球菌血清与丹麦国家血清研究所的产品比较 ,均有很好的特异性。结论　制备工艺稳定可靠 ,血清凝集效价高 ,特异性好 ,检测多糖的灵敏度高 ,经本实验室使用 。Objective To prepare type 9V diagnostic pneumococcal anti sera,eatablish one thechnological process and go on the working for the idenitification of pneumococcal capsular polysaccharide vaccine.Methods Immunizing rabbits with pneumococcal strains.Results The efficiency of agglutination of this serum was the 320 times with the reaction of the strains,and the 16 times with the reaction of the polysaccharide antigen.The determining sensityivity of the polysaccharide antigen was the value of ≥25 μg/ml with the double immunediffusion of this serum.The efficiency of this serum was the 2 times with the polysaccharide antigen reaction on the double immunediffusion.This serum has no the cross reaction with other type serum and with a good specificity.Conclusions This thechnological process was the stableand reliable for praparion of the serum.This serum has a high efficiency of agglutination and a good sensitivity in the determining sensitivity of the polysaccharide antigen and with a good specificity.
