摘  要:目的 探讨小鼠表皮角质形成细胞(KC)中Ca^(2+)对组织型纤溶酶原激活剂(tPA)表达的调节作用及其与分化的关系。方法采用免疫组化及原位杂交技术定性、定量检测低Ca^(2+)( 0.05 mmol/L)及高Ca^(2+)(1.5 mmol/L)浓度对tPA、tPA mRNA的表达及表皮KC分化的影响。结果与未经Ca^(2+)处理的对照相比,低Ca^(2+)浓度下培养的表皮KC中,tPA mRNA及tPA量均增加(P< 0.05),KC分化标记物K10抗原表达为弱阳性,KC胞体较小,细胞形态趋于圆形;而高Ca^(2+)浓度下的培养表皮KC中,tPA mRNA及tPA量显著增加(P< 0.001),K10抗原呈强阳性表达,KC胞体增大,形态为典型的多角形。结论Ca^(2+)浓度的增高促进了tPA mRNA及tPA的表达,并与KC分化有关。Objective To explore the regulation of calcium on the expression of tissue plasminogen activator (tPA)and its relationship with cell differentiation in mouse epidermal keratinocyte(KC).Methods In situ hybridization and immunohistochemistry were employed to study the effects of low and high concentra-tions of calcium on tPA expression and epidermal KC differentiation quantitatively and qualitatively. Re- sults When the epidermal KCs were incubated with low concentration of calcium, the expression of tPA mRNA and tPA were significantly increased compared with the control(P< 0.05), but the antigen of differentiation marker K10 expressed weakly, and the small and approximately round cells could be seen; When incubated with high concentration of calcium, the expression of tPA mRNA and tPA in KC were significantly increased very much compared with the control (P< 0.001), while the K10 antigen expressed strongly, the horny cells became much bigger and flatter. Conclusions The expressions of tPA mRNA and tPA were enhanced and related to KC differentiation by elevating calcium concentration .
