糖尿病母亲和新生儿血清瘦素水平及胎盘瘦素表达的研究

摘　　要：目的　比较正常和糖尿病母儿血瘦素水平及胎盘瘦素的表达。方法　采用放射免疫法测定 2 1例糖尿病母儿和 32例正常母儿血瘦素及胰岛素水平 ,应用逆转录多聚酶链反应 (RT PCR)技术检测胎盘瘦素mRNA表达。结果　糖尿病组母亲血清瘦素和胰岛素水平与正常对照组无显著差异 ,而糖尿病组新生儿脐血瘦素 (P <0 0 0 1)和胰岛素 (P <0 0 0 1)水平明显高于正常对照组。糖尿病母亲胎盘瘦素mRNA表达显著高于正常对照组 (P <0 0 0 1)。结论　糖尿病母亲所生新生儿有较高的脐血瘦素水平 ,且糖尿病母亲胎盘瘦素表达水平高于正常对照 。Objective To compare the expression of leptin in placenta and the level of serum leptin in diabetic mothers and neonates with normal control.Methods The concentrations of leptin were measured by the method of radioimmunoassay in 21 diabetic mothers and the neonates,as well as in 32 healthy mothers and the term newborns.The expression of leptin in placenta was also determined by RT PCR techniques.Results There was no difference of serum leptin and insulin levels between diabetic mothers and normal controls.The neonates of GDM(gestational diabetes mellitus) mothers had higher concentrations of leptin( P <0 001)and insulin ( P <0 001) in cord blood than the normal controls.Upregulation of the expression of leptin in placenta was also observed in GDM mothers.Conclusion The neonates of GDM mothers have higher level of serum leptin,and GDM mothers have increased expression of leptin in placenta.The results indicated that abnormal expression of leptin in placenta of GDM mothers may be one of the reasons of macrosomic infant.
