摘 要:目的 研究腺苷对猪急性出血坏死性胰腺炎 (AHNP)肠道屏障功能保护作用的机制。方法 12只成年小型猪经ANHP成模后随机均分成对照组和腺苷治疗组 (简称腺苷组 ) ,比较两组动物在AHNP诱发前后不同时相点肠道血流量、肠道通透性、门静脉血细菌和内毒素的变化。结果 (1)腺苷组在AHNP成模后 8,2 4h和第 7d时肠道血流量显著高于对照组 (P <0 .0 1或P <0 .0 5 ) ;(2 )腺苷组成模后 8,2 4h和第 3d肠道通透性显著低于对照组 (P <0 .0 5或 P <0 .0 1) ;(3)对照组门静脉血细菌和内毒素明显增加 ,腺苷组门静脉血细菌数在AHNP诱导后第 2 4h和第 3d较对照组显著下降 (P <0 .0 5或P <0 .0 1) ;门静脉血内毒素含量显著低于对照组各时相点 (P <0 .0 1或 P <0 .0 0 1)。结论 腺苷能显著增加肠道血流量 ;腺苷可以明显减轻肠粘膜损害 ,保护肠道屏障功能 ,腺苷的这种作用可能是通过增加肠道血流量而实现的 ;Objective To study the protective mechanism of adenosine on intestinal barrier function in acute hemorragic necrotizing pancreatitis (AHNP) pigs; Methods Twelve small pigs were equally devided into two groups randomly after the AHNP model sitted up: (1) AHNP controll group (group A) and adenosine treated group (group B). The intestinal blood flow, intestinal permeability, bacterial culture and endotoxin in portal blood were compared between the two groups in pre and post AHNP. Results (1)The intestinal blood flow was dramatically decreased in group A, and much higher in group B than that in group A at the 8 h, 24 hour and day 7 after ANHP model was setted up (P<0.01, P<0.05). (2) The gut became more permeable after AHNP was induced in both groups; the gut permeability in group A was rather higher than that in group B (P<0.05, P<0.001). (3) Intestinal bacteria and endotoxin in portal blood dramatically increased in group A. Portal blood bacteria was much less in group B than that in group A at 24 hour and 3 days respectively (P<0.05,P<0.01), and portal blood endotoxin in group B was much less than that in guoup A at all the time points respectively (P>0.001,P<0.001) except pre-AHNP. ConclusionsThe results suggest that: (1) Adenosine can obviously increase AHNP pigs' intestinal blood flow. (2) Adenosine can alleviate intestinal mucosal damage and protect gut barrier function, which may be realized through its effect on intestinal blood flow. (3)Adenosine can alleviate the translocation of intestinal bacteria and endotoxin.
