摘 要:目的 建立适用于实验的免疫鼠模型 ,并确定建立免疫鼠模型的最佳剂量和时相。方法 用免疫血清学测定和光镜及电镜方法研究经口伤寒杆菌 (salmonellatyphi,ST)和柯萨奇病毒 (coxsackievirus ,CV)诱导的小鼠肠道粘膜免疫应答。结果 ST和CV均可引起肠道粘膜免疫应答。ST经口灌注造模较为适宜的剂量为 10 3 个菌 /ml,CV的剂量为 10 -12 单位 /ml,两者第 2次灌注后 7天为最佳时相。此时小鼠回肠和回肠集合淋巴小结及肠系膜淋巴结内淋巴细胞明显增多 ,集合淋巴小结明显增大 ,淋巴细胞增殖活跃 ,且未发生病理改变。结论 ST和CV是研究肠道粘膜免疫建立免疫鼠模型较理想的实验工具。Objective To establish the two immune mouse models which was suitable for the research of experiment induced by salmonella typhi(ST) or coxsackie virus(CV) . The best treated dose and time course of the models were determined. Methods The mucosal immune response in mouse Gut by ST or CV was studied by immune serology test, light and electron microscopy. Results ST or CV could induce the mucosal immune response in mouse Gut. In ST model, the dose of ST 10 8 bacteria/ml was most responsive, but in CV model, the dose of CV was 10 -12 unit/ml. The best time course of the two models was the 7th day after the second treatment. The experiment showed that the number of lymphocytes in ileum, Peyer′s patch and mesenteric lymph node greatly increased. Peyer′s patch increased more obviously by in size and number, and was active in growth. The changes of pathology did not appear in our experiment. Conclusion ST or CV is more suitable to establish immune mouse model for the experiment research of the mucosal immunity.
