摘 要:目的 研究周围神经提取液对失神经骨骼肌、损伤恢复期周围神经和骨骼肌的作用。方法 将Wistar大鼠 48只随机分为对照组和实验组 ,将羊周围神经提取液注射于大鼠离断神经和修复神经损伤后的趾长伸肌中 ,检测其周围神经和骨骼肌的组织学指标。结果 失神经实验中 ,实验组中肌纤维平均面积、肌纤维平均直径、肌肉内肌纤维平均面积百分比、趾长伸肌指数均优于对照组。损伤恢复期实验中 ,实验组各指标均优于对照组。其中坐骨神经提取液组和粗提周围神经提取液组趾长伸肌指数为 (68.2 4± 9.54 ) %、(66.1 5± 4.46) % ,较生理盐水组 (61 .3 1± 4.52 ) %差异有显著性 (P <0 .0 5) ;腓总神经传导速度 (3 .3 9± 0 .91 )m/s、(5.3 1± 2 .72 )m/s[生理盐水组为(0 .86± 0 .77)m/s];有髓神经面积恢复率为 (1 6.49± 3 .49) μm2 、(1 6.0 2± 4.1 5) μm2 [生理盐水组为 (6.40± 3 .49) μm2 ];骨骼肌纤维平均面积为 (1 3 1 1± 42 9) μm2 、(1 2 56± 3 63 ) μm2 [生理盐水组为(598± 1 95) μm2 ]。结论 周围神经中含有缓解失神经肌肉萎缩的骨骼肌营养物质 ,周围神经提取液缓解了失神经骨骼肌的萎缩 ,促进了修复后神经和骨骼肌的恢复。Objective To study the effects of peripheral nerve extracts on denervated skeletal muscle, injured nerve and injured skeletal muscle.Methods Sciatic nerve extract of sheep was injected into the denervated and repaired extensor digital longus muscle (EDL). After 2 weeks in denervated EDL part, the EDL index and muscular histologic indexes were analyzed. After 4 weeks in the repaired nerve part, the EDL index and muscular histologic, electric physiological, peripheral nerve histologic indexes were analyzed.Results In the denervated EDL part all indexes of experiment groups were better than in the control group. In the repaired nerve part all indexes of experiment groups were better than in the control group. The extracts of peripheral nerve could promote the injured nerve recovery and alleviate the atrophy of skeletal muscle. The EDL index was (68.24±9.54)% by the sciatic extract, (66.15±4.46)% by the pure extract and (61.31±4.52)% by normal saline, respectively. The conduct velocity of nervus peroneus communis in the presence of sciatic extract was (3.39±0.91) m/s, in pure extract (5.31±2.72) m/s, in normal saline was (0.86±0.77) m/s, respectively. The recovery rate of myelinated nerve in the presence of sciatic extract was (16.49±3.49) μm 2, in pure extract was (16.02± 4.15) ?μm 2, in normal saline was(6.40±3.49)μm 2, respectively.In the presence of sciatic extract the muscle fiber area was(1?311±429)μm 2,in pure extract was (1?256±363)μm 2,in normal saline was (598±195)μm 2,respectively.Conclusion Peripheral nerve extract can alleviate denervated skeletal muscular atrophy and promote recovery of the injured peripheral nerves and skeletal muscle in vivo.
