摘 要:目的 研究半导体激光对血管内皮细胞增殖的影响以及内皮细胞的细胞间粘附分子-1(ICAM-1)表达的变化。方法 应用半导体激光、低强度氦氖激光辐照体外培养的血管内皮细胞,采用噻唑蓝(MTT)比色法研究内皮细胞增殖情况,用免疫组化方法研究其ICAM-l表达的变化。结果 半导体激光辐照内皮细胞后可促进细胞增殖,增殖效果要好于低强度氦氖激光;辐照结束后的48、72和96h发现同一幅照时间不同功率的两种激光,以 2mw促进内皮细胞增殖的效果最好,3 5 mW次之,0.7 mW最差;同一功率不同辐照时间的两种激光,以辐照 30 min促进内皮细胞增殖效果较好,20 min次之,10 min最差。两种激光辐照内皮细胞1次、2次和3次后,观察其ICAM-1的表达有下降趋势。结论 半导体激光在辐照血管内皮细胞促其增殖方面优于氦氖激光;同一辐照时间的两种激光,以2mW促进内皮细胞增殖的效果最好;问一功率的两种激光以辐照 30 min促进内皮细胞增殖效果较好;低强度激光对内皮细胞 ICAM-1表达有抑制作用。Objective To observe endothelial cell proliferation and variation of the expression of ICAM- 1 by using semiconductor laser irradiation on cultured endothelial cells. Methods Cultured rat arterial endothelial cells were treated with two kinds of low intensity laser (semiconductor laser, helium-neon laser) irradiation on cell proliferation and the expression of ICAM-1. Results A significant stimulative effect on cell proliferation was found statistically in semiconductor laser group and it is better than helium-neon laser group; According to the stimulative effects on cell proliferation of these two kinds of lasers with the same irradiation time, the group of 2 mW is better than the group of 3.5 mW and the later is better than the group of 0.7 mW; According to the stimulating effects on cell proliferation of these two kinds of lasers with the same energy, 30 minute group is better than 20 minute group and the later is better than 10 minute group; The expression of WAM-1 on cultured endothelial cells has a decreasing trend with the increase of irradiationg times. Conclusion Semiconductor laser irradiation has a more effective stimulating effect on cultured endothelial cell proliferation comparing with helium- neon laser;With the same irradiating period of time, 2 mW is the best choice for cell proliferation;With the same irradiating energy, 30 minutes is the best irradiating period of time for cell proliferation. Different kinds of low intensity lasers can restrain the expression of ICAM-1 of endothelial cells.
