摘  要:目的 探讨重组人生长激素 (rhGH)、胰岛素样生长因子 1(IGF 1)对烧伤小鼠巨噬细胞 (M)功能的影响。 方法 采用逆转录聚合酶链反应 (RT PCR)和酶联免疫反应方法 (ELISA) ,观察在体和离体应用rhGH、IGF 1对M表达CD14和分泌细胞因子的影响。 结果 烧伤后应用rhGH及IGF 1均能增加小鼠腹腔MCD14mRNA的转录和细胞因子的分泌 ,但两因素之间没有叠加作用。rhGH(40 μg/ml)和IGF 1各浓度均可促使培养的小鼠M分泌TNF、IL 6增加。  结论 rhGH可能由IGF 1介导 ,通过增加MCD14的表达激活M ,使其更多的分泌细胞因子 ,有利于增强免疫功能。烧伤后应用 0 2mg·kg-1·d-1剂量的rhGH ,无过度激活M的作用。Objective To investigate the effects of rhGH and IGF 1 on the CD14 mRNA expression and cytokine secretion of peritoneal macrophages (M) in scalded rats. Methods Reverse transcription polymerase chain reaction (RT PCR) and enzyme linked immunosordent assay(ELISA) were employed to observe the postburn changes of CD14 expression and cytokines secretion of the M treated in vivo and in vitro by rhGH and IGF 1. Results It was demonstrated that in vivo postburn application of rhGH and IGF 1 could enhance the transcription of CD14 mRNA and the secretion of cytokines from the peritoneal M in mice.But it seemed that there was no additive effect between the two factors.Additionally,rhGH(at 40 ng/ml) and IGF 1 (at all tested concentrations) could activate the secretion of TNF and IL 6 from cultured mice M in vitro. Conclusion Postburn application of rhGH might activate M by IGF 1 induction,enhancing M CD14 expession,which promoted more secretion of cytokines,and promote the enhancement of immune function. [
