抗肾综合征出血热病毒和抗辣根过氧化物酶的双特异性单克隆抗体的建立

摘　　要：成功地获得了既抗肾综合症出血热病毒,又抗辣根过氧化物酶的双特异性单克隆抗体(简称抗HFRSV-抗HRP BsMcAb)。方法是,先将分泌抗HFRSV McAb的杂交瘤A25-1细胞用杂氮鸟嘌呤进行诱变试验,建立了分泌抗HFRSV McAb的HGPRT细胞株(即HAT敏感型细胞株)251AGD6。用此细胞与HRP免疫的Balb/c小鼠脾细胞再次融合,得到分泌抗HFRSV-抗HRP的BsMcAb杂交杂交瘤(Hybrid Hybridoma)261-HRPB10细胞株。用双抗原免疫斑点试验和双抗原ELISA试验测定251-HRPB10腹水BsMcAb,滴度均为1000;用IFA测定单向抗HFRSV抗体滴度为10000;用免疫斑点法和ELISA测定单向抗HRP抗体滴度,均为100000。杂交杂交瘤的染色体数目在150条左右,证实是三细胞两次杂交瘤。An hybrid hybridom, 251-HRPB10, which secretes anti-hemorrhagic fever with renal syndrome virus (HFRSV) and anti-horseradish peroxi- dase (HRP) bispecific monoclonal antibody (BsMcAb),was obtained.The titer determined for BsMcAb by bi-antigen dot immunobinding assay and bi-antigen sandwich. ELISA was 1 : 1 ,000. The titer determined for anti-HFRSV McAb by IFA was. 1:10,000. The titer determining for anti-HRP McAb by dot immunobinding assay and ELISA was 1:100,000. The chromosome number of the hybrid hybridoma 251-HRPB10 was about 150. Three peaks were found in 251-HRPB10 ascites fluids analysed by DEAE-cellulese. The first, second and third peaks were anti-HFRSV McAb anti-HFRSV-HRP BsMcAb and anti-HRP McAb respectively, the second peak being the highest. Production of BsMcAb against viruses was very important for the therapy and diagnosis of virus diseases.
