摘 要:目的 :观察系统性红斑狼疮 (SLE)患者外周血单个核细胞 (PBMC)NF AT和AP 1活性 ,并进一步探讨其临床意义。方法 :NF AT和AP 1活性检测采用电泳迁移率改变法 (EMSA)。结果 :①活动期 ( 1 9例 )和缓解期 ( 1 3例 )SLE患者PBMCNF AT活性均显著高于正常对照组 ( P <0 0 5) ;活动期SLE显著高于缓解期 ( P <0 0 5)。②活动期SLE组PBMCAP 1活性均显著低于正常对照组 (P <0 0 1 ) ;缓解期SLE与正常对照组无明显差别 (P >0 0 5)。③血清抗dsDNA抗体阳性SLE患者 ( 2 1例 )NF AT明显高于抗dsDNA抗体阴性组 ( 1 1例 ) ( P <0 0 5) ,而两组患者AP 1活性无显著差异 ( P >0 0 5)。④SLE患者PB MCNF AT活性与CRP和SLEDAI呈正相关关系 ,与ESR、ANA、C3无相关关系 ;AP 1与上述各临床指标无相关关系。结论 :SLE外周血单个细胞中存在介导细胞内信号转导的转录因子NF AT和AP 1表达异常 ,这可能与SLE的发病机制有关 ;NF AT可能与SLE的疾病活性有关。Objective:To detect the activation of NF AT and AP 1 in peripheral blood mononuclear cells(PBMCs) in patients with systemic lupus erythematosus and investigate their relationship with serum ANA,anti dsDNA antibody,ESR,CRP and C 3.Methods:Activation of NF AT and AP 1 were detected by electrophoretic mobility shift assay(EMSA).Results:①Activation of NF AT in PBMC in active and inactive SLE patients was increased when compared with normal controls,respectively,and the activation of NF AT in active patients was higher than that of inactive ones.②Elevated activation of AP 1 in PBMC in active SLE patients was found as compared with normal controls,and there was no significant difference in activation of AP 1 between inactive patients and normal control.③Activation of NF AT in SLE patients with positive anti dsDNA antibody was higher than that of patients with negative anti dsDNA antibody,and there was not different positively in AP 1 activation between above two patient groups.④Activation of NF AT in PBMC was related positively with CRP and SLEDAI,respectively,but no correlation with ESR,ANA,C 3.There was no relation between the activation of AP 1 and above clinical parameters.Conclusion:The abnormal expression of NF AT and AP 1 indicates disturbed intracellular signaling in SLE,which is suggested that alternations in activation of transcription factors may contribute to the pathogenesis of SLE.Activation of NF AT may be used as a predicator of SLE disease activity.
