银杏内酯A和银杏内酯B在健康人体内的药动学的实验研究

摘　　要：目的　用本实验室建立的测定血液中内酯浓度的方法 ,对银杏内酯A(ginkgolideA ,GA)和银杏内酯B(ginkgolideB ,GB)在 10例健康受试者体内的药动学进行了研究。方法　 10例健康受试者 ,静脉滴注银杏叶提取物注射液 (舒血宁注射液 ) ,于给药后不同的时间取血 ,用气相 质谱联用法测定血药浓度 ,并计算GA和GB的药动学参数。结果　健康受试者静脉点滴GA和GB后的代谢均符合二房室模型 ,t1/ 2 分别为 3.73,4.2 5h。AUC值分别为 6 830 5 .10 ,19714.6 9ng·min·mL-1。结论　GCOBJECTIVE To study the pharmacokinetics of ginkg ol ide A(GA) and ginkgolide B (GB).METHODS The Ginkgo biloba extract(GBE) injection was administered by intravenous infusion in 10 volunte ers and GC-MS was used to determine the concentrations of GA and GB in plasma. RESULTS The dispositions of both GA and GB were conformed to two-compartment model.t 1/2 of GA and GB were 3.73 h and 4.25 h,respe ctively.The AUC of GA and GB were 68 305.10 ng·min·mL -1 and 19 714.6 9 ng·min·mL -1 respectively.CONCLUSION The GC-MS ass ay was sensitive for the determination of GA and GB in human blood.The pharmacok inetic parameters of GA and GB from 10 volunteers were similar to those reported .
