反相高效液相色谱法测定远志中远志皂苷元的含量

摘　　要：目的 :建立反相高效液相色谱法测定远志中远志皂苷元的含量。方法 :采用反相高效液相色谱法。色谱柱为ODS柱(2 5 0mm× 4 6mm ,5 μm) ,流动相为乙腈 -水 - 36 %乙酸 (4 9∶5 1∶0 5 ) ,检测波长为 2 15nm ,流速为 1 0mL·min-1。结果 :线性范围是 42～ 5 2 4μg·mL-1,r =1 0 0 0。理论塔板数为 6 143。测定了我国远志主产区山西 6个产地的远志中远志皂苷元的含量。其含量范围为 1 47%～ 1 5 9%。结论 :本方法灵敏可靠 ,可以用来控制远志的质量。Objective:To establish a RP-HPLC method for determining senegenin in Radix Polygalae.Methods:The content of senegenin was determined by RP-HPLC. ODS column(250 mm×4 6 mm,5 μm) was used with a mobile phase of acetonitrile-water-36% acetic acid (49∶51∶0 5) and detected at 215 nm.The flow rate was 1 0 mL·min -1 .Results:The range of linearity is from 42 μg·mL -1 to 524 μg·mL -1 ( r =1 000).The theoretical plate number is 6 143.The content of senegenin in Radix Polygalae from six districts of Shanxi province was measured with the above method.The range of contents was 1 47%～1 59%.Conclusion:The established method is sensitive,reliable and suitable for the quality control of Radix Polygalae.
