摘  要:利用PCR和RFLP技术分析了小麦赤霉酸反应不敏感的矮秆基因Rht3的近等基因系及其分离群体。RAPD分析从 31 0条随机引物中 ,筛选出 3个引物可以在高矮亲本中稳定地扩增出多态带 ,连锁分析表明仅S1 0 60 190 0 和S1 0 60 2 0 0 0 扩增片段与Rht3连锁 ,遗传距离分别为7.1cM和 9.2cM。RFLP分析选用了主要位于第 4部分同源群短臂上的 5 3个探针 ,其中Xpsr5 84、XksuF8和Xcdo383个探针在高矮亲本中揭示出多态性 ,连锁分析表明仅Xpsr5 84与Rht3基因连锁 ,遗传距离为 8.0cM。The dwarf gene Rht3 derived from Tom Thu mb variety, a Tibetan wheat of China, is a dominant gene with the insensitivity to gibberellic acid. Rht3 shows to depress α-amylase activity and hence enchance the resistance to preharvest sprouting mainly through its effect on the expression of α-Amy1. Near isogenic lines with Rht3 and their segrega ting populat ion were analyzed by PCR and RFLP techonology. In RAPD analysis, out of 310 rand om primers (10bp) screened, only three primers of UBC389, OPV-06 and S1060 revea led polymorphisms in NIL from 310 random primers. Fragments S1060 1900 and S1060 2000 amplified by primer S1060 were shown to be linked to Rht 3 wi th a genetic distance 7.1cM and 9.2cM. Fifty-three probes specific for short ar ms of homoeologous group 4 were screened in RFLP analysis. Xpsr584, Xksu F8 a nd Xcdo38 showed polymorphisms between the NIL. The linkage analysis indicat eded that Xpsr584 was linked to Rht3 with a genetic di stance 8.0cM.
