广东汉族人血管紧张素Ⅰ转换酶基因I/D多态性与冠心病的关系

摘  要:目的 探讨ACE基因I/D多态性是否与广东人群患冠心病 (CAD)有关。方法 用聚合酶链反应 (PCR)技术分析了 72例广东汉族人CAD患者的血管紧张素Ⅰ转换酶 (ACE)基因插入 /缺失 (I/D)多态性 ,并与 186例正常广东汉族大学生ACE基因频率相比较。同时用比色法测定外周血中ACE活性。结果 CAD患者Ⅱ基因型频率显著低于正常对照组 ( 35 %vs 49% ) ,D基因携带者 (DD +ID)明显高于对照组 ,且DD基因型和ID基因型患者发病年龄明显提前。广东汉族大学生不同基因型血浆ACE活性明显不同 ,DD型人最高 ,ID型次之 ,Ⅱ型最低。结论 ACE基因I/D多态性与广东汉族人血浆ACE活性有关 ,D等位基因携带者易患CAD。Objective To study the association between angiotensin Ⅰ-converting enzyme (ACE) gene I/D polymorphism and CAD in Guangdong. Methods\ ACE gene I/D polymorphism and plasma ACE activity were studied in 72 Guangdong individuals with coronary artery disease (CAD) and compared to 186 unrelated normal Guangdong. Results\ The Ⅱ genotype frequency was significantly lower (35% vs 49%) and the frequency of D allele gene carriers(DD+ID) was higher. In CAD patients DD genotype patients had the lowest onset age and Ⅱ genotype had the highest. In the normal Guangdong Han individuals, plasma ACE activity was highest in DD genotype, followed by ID and then by Ⅱ genotype. Conclusion\ ACE gene I/D polymorphism was associated with plasma ACE level, and it may be a risk factor for CAD in Guangdong Han individuals. Subjects with D allele seem to be susceptible to CAD.
