摘  要:目的探讨膜结合表达对基因免疫的影响。方法流感病毒跨膜蛋白基因修饰带 HBV pre C分泌型信号肽的HCV C6 9基因 ,构建 pc6 9TM质粒 ;35S-甲硫氨酸代谢标记和免疫沉淀检测质粒体外瞬时表达蛋白 ;肌肉免疫 Balb/c小鼠。结果 pc6 9TM基因表达出的蛋白存在于膜上。免疫鼠淋巴细胞对特异性抗原刺激的增殖能力 pc6 9TM较分泌表达型质粒pc6 9显著增强 (P=0 .0 3) ,而 2种质粒诱导小鼠产生抗体效价差异不显著 (P=0 .76 5 )。结论膜结合表达能增强 HCV C6ObjectiveIn order to design suitable vectors as DNA vaccines against infections with the HCV, we investigated if membrane retention of the viral antigen has an influence on the humoral immune response and proliferative activity Methods pc69TM plasmid was constructed, in which HCV C69 gene with HBV preC signal peptide DNA sequence was adorned by transmembrane protein (TM) gene of influenza virus Products of transient expression in COS 7 cells were de tected by labeling with 35 S Met and immuno precipitation Balb/c mice were given injections of the plasmids intramuscularly Results HCV C69 protein was intracellularly retained in membrane The plasmid containing pc69TM can induce significantly higher lympho proliferative activity than that of pc69( P =0 03),but the antibody response to pc69TM and pc69 is not significantly different( P =0 765) Conclusion lympho proliferative activity to HCV C antigen can be enhanced by adornment of TM
