益气化瘀煎剂抗大鼠胃溃疡作用及其机制的探讨

摘　　要：目的 :研究益气化瘀煎剂对大鼠实验性胃溃疡的防治作用 ,并对其机制进行探讨。方法 :观察预先应用中药对应激、乙醇、消炎痛、幽门结扎所诱发的胃粘膜损伤的影响 ;并观察连续应用中药 8、18和 2 6d后 ,醋酸性胃粘膜损伤的愈合情况 ;检测胃壁结合粘液、胃组织DNA、胃粘膜超氧化物歧化酶 (SOD)、丙二醛 (MDA)及前列腺素E2 (PGE2 )的含量。结果 :益气化瘀煎剂显著抑制应激、乙醇及消炎痛性胃粘膜损伤 (P <0 .0 5 ,<0 0 1) ,并具有剂量依赖关系 ;加速胃溃疡的愈合 ,其疗效与雷尼替丁接近。同时 ,增加胃粘膜组织DNA、SOD及PGE2 的含量 ,减低MDA的生成。结论 :益气化瘀煎剂具有较强防治胃溃疡的作用 ,其机制可能主要是增强胃粘膜细胞保护作用。Objective:The mechanisms of antiulcer effects of decoction Yiqi Huayu were investigated.Methods:By measure the content of the combination of gastric wall with mucus,DNA,SOD,MDA and PGE 2 we observed that the effects of precedingly offering Chinese drugs(20?10 or 5 g/kg×7 d,ig) on the gastric ulcers in rats induced by stress,alcohol,Indomethacin, and pyloric ligation, and that the cure of acetic acid induced gastric ulcer after continuously using these drugs 8,18,and 26 days.Results:By incresing the combination of gastric wall with mucus,DNA,SOD,and PGE 2 in gastric tissue,and decreasing the generation of MDA, decoction Yiqi Huayu dose dependently inhibited the formation of the gastric ulcer induced by stress,alcohol and Indomethacin respectively,and also accelerated the healing of gastric ulcer induced by acetic acid.The effects of these drugs were similar to that of Ranitidine.Conclusion:It is suggested that the effective antiulcer effects of decoction Yiqi Huayu may mainly relate to increasing the activity of gastric mucosal cytoprotection.
