摘  要:研究了可用于免疫学、分子医学、分子生物学、临床检验 ,也可用于药物筛选和开发新药的表面等离子体共振免疫传感器。这种传感器以横向塞曼激光器为光源 ,同一光束中包含正交的 P偏振光和 S偏振光 ,完全满足共光路原则。相位检测能避免电路的直流漂移影响 ,提高测量精度。同时 ,在研究了相位变化与金属膜的材料和厚度关系后 ,优化有关设计参数 ,构建了一个实验装置 ,并进行了蓖麻毒素蛋白与其抗体相互作用的动力学研究。实验结果表明 ,这种传感器灵敏度高 。A surface plasmon resonance immunosensor based on optical heterodyne phase detection is described. It can be used in immunology, molecular medicine, molecular biology and clinical tests as well as in drug screening and development of new drugs. This sensor employs a transverse Zeeman laser as the light source, and the same beam contains both orthogonal polarized lights P and S with complete path sharing. For phase detection, the measurement accuracy is improved by replacing DC detection of luminous intensity with AC detection to avoid the impact of shift in the case of DC detection. Theoretical analysis indicates that phase change relies strongly on the thickness of the sensing membrane and the material and thickness of the metallic membrane. An experimental apparatus was constructed with optimized design parameters. A study of the dynamic interaction between ricin and its antibody was conducted. Experimental results show that this sensor has good performance in sensitivity, stability and reliability.
