摘 要:利用随机扩增多态性 DNA(RAPD)技术对 7株杜氏藻 ,即 :D.salina、D.bardawil、D.minu-ta、D.bioculata、D.parva、D.peircei和 D.p rimolecta进行了遗传多态性分析 .用 30个 10聚随机引物进行多态性扩增和筛选 ,其中 2 3个引物 (76.67% )可获得清晰的多态性条带 .选择其中能够得到清晰、重复性良好的 18个引物共扩增产生 15 0条可区分的 DNA条带 .根据种间 Nei遗传相似系数计算分析 ,构建杜氏藻属部分种的聚类分析图 .分析结果表明 :D.salina、D.bardawil和 D.minuta聚为一类 ,D.bioculata、D.parva、D.peircei和 D.primolecta聚为另一类 .由于两类之间存在较远的亲缘关系 ,把这两类杜氏藻称为两个种可能更合适 .Seven species of Dunaliella were analyzed by using random amplified polymorphic DNA (RAPD) techniques. Thirty decamer oligonucleotide random primers have been used for polymorphic selection. Of the 30 primers, 23 (76.67%) were found to produce polymorphic products. A total of 150 distinctive bands amplified from 18 primers were selected for further Nei's similarity coefficients analysis and to construct dendrogram by using UPGMA in PHLIPS programs. The results mentioned above showed that: D.salina, D.bardawil and D.minuta were clustered into one group, while D.bioculata, D.parva, D.peircei and D.primolecta were clustered in another one. Because of finding far away relationship between the two groups of Dunaliella, as our opinion, it may be more reasonable for regarding the two groups as two species.
