摘 要:用放射配基测定法探讨人和大鼠血淋巴细胞β受体与[~3H]DHA结合的饱和性质。试验表明:人及大鼠血淋巴细胞β受体存在高亲和力低容量及低亲和力高容量两个结合部位,在测定有生理活性的高亲和力低容量位点时,正常人完整血淋巴细胞β受体密度(Bmax)为548.64±79.71fmol/10~7细胞,平衡解离常数(Kd)为6.80±1.40nM。正常SD大鼠完整血淋巴细胞β受体Bmax为285.21±32.71 fmol/10~7细胞,Kd为5.94±1.47nM。The two binding sites of β-adrenergic receptors on circulating intact lymph-ocytes (CIL) were not recognized by most of the reseachers who studied the characters ofβ-receptors on CIL. We used [~3H]dihydroalprenolol to study some kinetic characteristic ofnormal SD rats and human beings. The data are represented as follows: β-receptors in humanbeings and SD rats show two binding sites: the high affinity low capacity site and lowaffinity high capacity site. In the experimental conditions we used, the B_(max) and the K_d^dvalues of high affinity low capacity site of β-receptor on CIL in human beings(n=12) anrats(n=10) were 584.64±79.71, 258.53±32.71 fmol/10~7 cell and 6.80±1.41, 5.94±1.477nM(Mean±SEM)respectively. For the first time, the presented data suggest that the highaffinity low capacity site on β-receptors on CIL is available and valuadle to be widely employedin the clinic and physiologic studies of β-receptors.
