摘　　要：本文探讨ABC-ELISA检测抗精子抗体的实验条件。结果提示:最适包盖精子浓度为10×10~9/ml;最适血清稀释度为1:400;甲醛与戊二醛固定效果无显著性差异(P】0.05),10％FCS-PBS封板并不显著降低非特异反应(P】0.05);批内和批间误差分别为2.1％～4.3％和6.4％～9.4％;比较检测男,女阳性血清灵敏度,AEC-ELISA较普通ELISA敏感4倍,较Franklin-Dukes试验敏感100倍;比较检测兔抗人精子免疫血清灵敏度,则分别敏感8倍和400倍。本法具有灵敏度高、特异性强和稳定性好等优点,作者认为,不仅可用于循环抗体的检测,而且在局部抗体检测上也具有应用前景。The experimental conditions of ABC-ELISA using for detecting the antisperm antibody were reported. It was found that the optimal concentration of the coating sperm was 10×10~6/ml and the optimal dilution of the serum was 1: 400, there was no significant difference in the fixative effects between formalin and glutaraldehyde (P>0.05). 10％ FCS-PBS sealing plate did not reduce the non-specific reaction significantly(P>0.05) .The repeatability showed that the CV of intraassay was 2.4％-4.3％ and CV of interassay was 6.4％-9.4％. ABC-ELASA was 4 times and 100 times as sensitive as standard ELISA and Franklin-Dukes respectively in the determination of both male and female positive samples, it was 8 times and 400 times as sensitive as the other two methods respectively in the determination of rabbit antihuman sperm antiserum. Because of the high sensitivity, specifity and stability, the authors hold that ABC-EL ISA is useful not only in detecting circulating antibody but also in the research on local antibody.
