大鼠心肌再灌注白介素-8的变化及激素干预的影响

摘　　要：目的　探讨大鼠心肌缺血 /再灌注期间血清IL 8和CK MB的变化及甲基强的松龙对其的影响。方法　将大鼠随机分成空白对照组、缺血 /再灌注对照组、空白治疗组、缺血 /再灌注治疗组四组 ,在心肌缺血前用甲基强的松龙 (30mg/kg)对治疗组大鼠预处理 ,测定缺血0、 0 5h和再灌注 1、 2、 3h血清中IL 8和CK MB的含量。结果　CK MB从再灌注 1h开始逐渐升高 ,且在 2、 3h升高明显 (P <0 0 1) ;而IL 8从再灌注 2h开始显著性逐渐升高 (P <0 0 1)。治疗组CK MB、IL 8表现为升高延迟 ,即CK MB和IL 8均延迟至再灌注 3h开始升高 ,同时在再灌 2、 3h的二者血清浓度与对照组比较均显著性降低 (P <0 0 5 )。另外IL 8、CK MB两者水平呈显著性正相关 (P <0 0 0 1)。结论　大鼠在心肌缺血 /再灌注中可以产生IL 8,并有逐渐递增的趋势 ,甲基强的松龙可以抑制IL 8的产生 ,减少心肌的炎性损害 。Objective In order to investigate changes of serum IL 8,CK KB and effection of methylprednisolone (MP) on IL 8 and CK KB during myocardial ischaemia and reperfusion in rats.Methods We randomly divided rats into four groups:control group of ischaemia and reperfusion,sham control group,MP treated group of ischaemia and reperfusion,sham treated group.We pretreated the rats of treated group with MP(30mg/kg) before ischaemia.Blood was taken for IL 8 and CK MB measurement after 0,1,2,3 hours of reperfusion post ischaemia.Results Serum concentration of CK MB started to increase gradually,and was significantly increased at 2 and 3 hours after reperfusion(P<0 01),while IL 8 was significantly and gradually increased from 2 hours (P<0 01).In the MP treated ischaemia and reperfusion group,the increase of concentration of CK MB and IL 8 was delayed and started to do so from 3 hours after reperfusion (P<0 05).At the same time,concentrations of CK MB and IL 8 were significantly lower than those of control group (P<0 05).In addition,expression of IL 8 positively correlated with CK MB(P<0 01).Conclusion During myocardial ischaemia and reperfusion,IL 8 was produced and increased gradually.MP can inhibit the production of IL 8 and reduce the myocardial inflammatory injury,and may play a role in protecting heart.
