摘  要:本文应用高压液相 电化学 (HPLC EC)技术对 5 2名男性不育患者精子DNA中 8 羟基脱氧鸟苷 (8 OHdG)进行测定 ,同时检测精液量、精子密度、精子活率、正常形态精子率 ,及头部精子畸形等精液参数 ,并进行相关性分析。结果显示 ,当精子DNA中 8 OHdG超过 10 .5 0时 ,其头部畸形率明显增加。精子DNA中 8 OHdG与精液量 (r =- 0 .4 7,P <0 .0 0 1)、正常形态精子率 (r=- 0 .36 ,P <0 .0 1)成一定的负相关 ,且与头部畸形精子率成正相关 (r =0 .6 9,P <0 .0 0 1)。结果提示 ,精子DNA中 8 OHdG与精液质量存在一定的相关性 ,表明精液质量的下降与精子DNA的损伤存在一定联系 ,而内源及外源性活性氧是可能的原因之一 ,其可能的机制尚需进一步研究。hydroxy 2' deoxyguanosine(8?OHdG),a typical form of DNA adducts,is a key molecular biomarker of DNA oxidative damage which can be measured by high peroformance liquid chromatography (HPLC) with electrochemical detector(EC).The aims of this study are to analyze the 8?OHdG level of sperm DNA and relationship with semen quality.In 52 male infertiles,8?OHdG level of sperm DNA were determined by HPLC?EC and semen quality were examined according to WHO guidelines for the examination of human semen.The relationships between 8?OHdG and semen quality were also analyzed.The mean 8?OHdG level of sperm DNA was 11.16 (8?OHdG/10 5 dG).It was determined that there were significantly negative relations between 8?OHdG level of sperm DNA and semen volume ( r=-0.47,P <0.001),sperm number ( r=-0.37,P <0.01),and normal morphology ( r=-0.036,P <0.01),respectively.The positive relation between 8?OHdG level and head defective sperm ( r=0.69,P <0.01) was also found.The study showed that there existed relationship between 8?OHdG level of sperm DNA and some semen parameters.These suggested that the poor semen quality was partly from the damage of sperm DNA owing to attacks of reactive oxygen species.However,the possible mechanism need to the further studied.
