摘 要:目的:探讨牵张刺激对心肌细胞NF-κB的激活作用及激活机制。材料与方法:在硅橡胶膜上培养心肌细胞,在无NAC和有NAC的条件下对细胞施加牵张刺激,用免疫组织化学方法观测NF-κB的激活情况。结果:牵张心肌细胞5小时后,NF-κB被显著激活;15mM N-乙酰半胱氨酸(NAC)阻断牵张诱导的NF-κB的激活。结论:牵张刺激能够诱导心肌细胞NF-κB的激活,活性氧介导了这种激活作用。purpose: To determine the activation of NF-κB and its mechanism. material and method: Cardiac myocyytes cultured on the silicon membrane were stretched in the presence of NAC and in the absence of NAC respectively,and then immunohistochemical method was applied to observe the activition of NF-κB in cardiac myocytes in different conditions. Results:NF-κB was activated significantly by stretching cardiac myocytes for five hours and stretch-induced activation of NF-κB was blocked by 15mM N-acetyl cysteine. Conclusion: The stimulation of stretch can induce the activation of NF-κB in cardiac myocytes, which is mediated by reactive oxygen.
