摘 要:利用小鼠胚胎卵巢无血清培养体系研究了 FSH、h CG对小鼠胚胎卵巢卵泡发育的作用。结果发现 ,(1) FSH处理组胚胎卵巢卵泡体外生长发育要优于 ITS对照组 ;体外培养 15 d后 ,FSH处理组胚胎卵巢睾酮分泌量显著增加 ;在整个培养过程中 ,FSH诱导胚胎卵巢分泌少量雌二醇。 (2 ) h CG对胚胎卵巢卵泡发育的作用是双相的。在培养早期 ,h CG处理组进入生长相的小卵泡显著多于 ITS对照组 ,相应地此时雌二醇水平较高 ;随着卵巢培养的继续 ,雌二醇水平显著降低 ,h CG处理组总卵泡数开始急剧下降 ,大量小卵泡闭缩退化 ;到培养结束时 ,处理组卵巢内小卵泡已所剩无几 ,但残存的生长卵泡卵母细胞平均直径仍显著大于 ITS对照组。此时伴随睾酮分泌量的增加 ,雌二醇的分泌量重新增加。 (3) FSH+h CG处理组卵泡发育类似 h CG处理组 ,体外培养 15 d后 ,FSH+h CG促进睾酮分泌 ,而雌二醇的分泌水平很低。结果提示 ,FSH虽然不是小鼠胚胎卵巢原始卵泡生长起始所必需 ,但对腔前卵泡的进一步发育及其雌二醇的分泌有促进作用 ;雌二醇可能是促性腺激素发挥促卵泡体外发育和存活作用所必需的 ;h CG可促进卵泡提前生长发育 。Our preliminary work has established a sera-free culture system for mouse fetal ovary.And then the effects of gonadotrophins on the mouse fetal ovarian follicle development and steroidogenesis was researched using the above system.The results were as following:(1)The growth and survives of fetal ovarian follicle in FSH group was significantly prior to that in ITS group.FSH enhanced the secretion of testosterone after 15 days culture.And estrodial could be detected in a low level throughout the culture period.(2)hCG had a biphasic role in the mouse fetal ovarian follicle development,which refers to that hCG promote the follicular development at the early culture period with a higher level of estrodial and induce the follicular atresia and degeneration at the mid-late culture period in company with a decrease of estrodial content.There were few follicles survived at the end of culture period,however the average diameter of five largest oocytes was still larger than that in ITS group.(3)The follicle growth and survival in FSH+hCG group was similar to that in hCG group.The level of testosterone increased after 15 days culture,however only a low level of estrodial induced.The results suggested:①FSH promotes the follicle development of mouse fetal ovarian and the secretion of estrodial,although FSH is not necessary for the initiation of primordial follicles;②Estrodial is able to energize the trophic role of FSH in the follicular development and survival in vitro;③hCG may accelerate the exploitation of the small follicle pool.
