苦马豆素-BSA对家兔的免疫原性研究

摘　　要：用完全福氏佐剂对 14只家兔进行基础免疫 10 d后 ,将家兔随机分成 2组 ,每组 7只 ,1组为苦马豆素 - BSA免疫组 ,另 1组为对照。首免时免疫组每只兔四足掌共注射 0 .8m L,两侧大腿内侧肌肉各注射 0 .3m L,两侧颈部皮下各注射 0 .3m L,免疫总剂量为 2 m L/只 ,抗原含量 2 m g/ m L。 5周后第 1次加强免疫 ,免疫组每只兔两侧大腿内侧肌肉各注射 0 .5 m L,背部皮下 4点注射 ,每点 0 .2 5 m L,免疫总剂量为 2 m L/只 ,抗原含量 3mg/ m L。 1周后试血。 10 d后第 2次加强免疫 ,免疫途径与第 1次加强免疫相同 ,免疫总剂量为 2 m L/只 ,抗原含量 6 mg/ m L。1周后试血。结果表明 ,苦马豆素 - BSA第 1次加强免疫 1周后 ,1～ 7号兔都产生了抗体 ,间接血凝试验测定的抗体滴度分别为 2 1 ,2 1 ,2 2 ,2 1 ,2 1 ,2 2和 2 2 ,间接 EL ISA试验测定的抗体滴度分别为 2 2 ,2 2 ,2 3,2 2 ,2 2 ,2 3和 2 3。第 2次加强免疫 1周后试血 ,1～ 7号兔抗体滴度都明显升高 ,间接血凝试验测定的抗体滴度分别为 2 6 ,2 6 ,2 8,2 7,2 7,2 8和 2 8,间接 EL ISA试验测定的抗体滴度分别为 2 7,2 7,2 9,2 8,2 8,2 9和 2 9。After basically immunized with complete Freunds adjuvant,14 rabbits were divided into 2 groups equally.Group one was immunized with swainsonine BSA,which was used as the vaccine after emulsifying it with complete Freunds adjuvant.The other was the control gruop.Immune procedure was as followings.The rabbit was injected 2 mL swainsonine BSA vaccine,containing 4 mg swainsonine BSA.0.8 mL was injected into 4 plantae,each fore plantae was 0.15 mL and hind plantae was 0.25 mL.0.6 mL was injected inside hind leg,with each leg 0.3 mL,i.m.0.6 mL was injected into neck,i.p.After 5 weeks the first intensive immunization was conducted,swainsonine BSA was dissolved in water for injection.2 mL of the solution,containing 6 mg swainsonine BSA,was injected to the rabbit.1 mL was injected inside hind leg,each leg 0.3 mL,i.m.1 mL was injected 4 points of back,0.25 mL in each point.Antibody was monitored after a week.The second intensive immunization was conducted after 10 days.Immune dose was 2 mL,containing 12 mg swainsonine BSA.Immune way was the same as the first intensive immunization.Antibody was monitored after a week.Immunoassay was conducted with indirect hemagglutination test and indirect ELISA test.After a week of the intensive immunization,all rabbits produced antibody.The titres,monitored by indirect hemagglutination test,were 2 1,2 1,2 2,2 1,2 1,2 2 and 2 2,and indirect ELISA test,were 2 2,2 2,2 3,2 2,2 2,2 3 and 2 3.After a week of the second intensive immunization,the titres,monitored by indirect hemagglutination test,were 2 6,2 6,2 8,2 7,2 7,2 8 and 2 8,and indirect ELISA test,were 2 7,2 7,2 9,2 8,2 8,2 9 and 2 9.
