摘  要:目的探讨无机砷对人皮肤成纤维细胞DNA的损伤特征。方法使用原代培养人皮肤成纤维细胞 ,应用单细胞凝胶电泳法(Singlecellgelelectrophoresisassay,SCGE)观察砷酸钠和亚砷酸钠对人皮肤成纤维细胞DNA的损伤特征。结果1~10μmol/L的砷酸钠染毒24h可诱发人皮肤成纤维细胞的DNA损伤 ,且具有剂量_效应关系。低浓度时以I级DNA链断裂为主。当浓度增高到10μmol/L时 ,II级DNA链断裂比例达50 % ,但未出现III级DNA链断裂。1μmol/L的亚砷酸钠染毒24h对人皮肤成纤维细胞DNA的损伤不明显。10μmol/L的亚砷酸钠可引起人皮肤成纤维细胞DNA的损伤 ,I,Ⅱ ,Ⅲ级DNA链断裂均有出现 ,且有较多的凋亡型DNA链断裂。结论砷酸钠主要引起一般型DNA损伤 ,而亚砷酸钠除引起一般型DNA链断裂外 ,还可引起凋亡型DNA链断裂。Objective To investigate the action of inorganic arsenic in the induction of DNA strand breaks in primary cultured human skin fibroblasts. Methods Sodium arsenate and sodium arsenite were used as test inorganic arsenics. DNA strand breaks were assessed by single cell gel electrophoresis assay (SCGE). Results Arsenate at 1~10 μmol/L dose_dependently induced DNA strand breaks in cells. Arsenate at lower concentrations induced mainly degree I DNA strand breaks, while the proportion of cells with degree Ⅱ DNA strand breaks increased to 50% when treated with 10μmol/L arsenate,but no cells with degree ⅢDNA strand breaks were observed.Cells treated with 1μmol/L arsenite showed no significant increase in DNA strand breaks. At the concentration of 10μmol/L, however, arsenite induced DNA strand breaks with different degrees, and the apoptotic type DNA strand breaks were the major type. Conclusion Sodium arsenate mainly induced general type DNA strand breaks and sodium arsenite induced apoptotic type,beside general type of DNA strand breaks in primary cultured human skin fibroblasts. This could be explained by their different reaction modes with DNA, and further studies were needed.
