摘  要:目的 :从鸡蛋中提取特异性抗体IgY ,并研究鸡蛋黄抗体免疫应答的持久性 ,以探索一种新的被动免疫防龋方法。方法 :以变形链球菌GTF高表达株S .mutansB2 9- 33及茸毛链球菌S .sobrinus 6 715免疫母鸡。采用硫酸铵沉淀法提取鸡蛋黄抗体 ,SDS -PAGE鉴定抗体性质 ,分光光度法检测蛋白质含量 ,间接酶联免疫吸附法 (ELISA)测定鸡血清及鸡蛋黄抗体效价滴度变化。结果 :SDS -PAGE鉴定证实 :用硫酸铵沉淀法可提取出鸡蛋黄抗体 ,其性质为IgG ,蛋白质含量约为每毫升 7.0 0mg鸡蛋黄。免疫应答持续时间超过 2 4周。结论 :鸡蛋黄抗体来源充足 ,抗体含量高 ,并能在一段较长时间内从免疫母鸡鸡蛋黄中获取 。AIM: The specificity of egg yolk antibody IgY against cariogenic bacteria and the protracted time of the antibody were detected to find a new passive immunization method. METHODS:50 hens were divided into two groups and injected with S.mutans B29-33 and S.sobrinus 6715 separately. The egg yolk antibody IgY purified by Sodium sulfate was determined with SDS-PAGE. The protein content was evaluated by absorption spectrometry method. Indirect ELISA was applied to evaluate the specific activity of IgY. RESULTS:SDS-PAGE showed that the purified egg yolk was IgG. The protein content was 7.00mg/ml. The specific activity lasted more than twenty-four weeks. CONCLUSION:Egg yolk antibody can be purified conveniently with high protein content. It has high specificity against cariogenic bacteria and can be obtained in a long time. So it can be used as a suitable source of antibody for passive immunization.
